چکیده
In this study, the antioxidative effect of melatonin was investigated in aging retinal pigment epithelial cells vitro. The objective study is to explore administration pharmacological doses for prevention symptoms.Specific concentrations (20 µM, 40 µM and 80 µM) were used treat hydrogen peroxide-induced flow cytometry employed examine cell apoptosis. A specific probe utilized detect intracellular reactive oxygen species concentration apoptosis-associated proteins detected real-time polymerase chain reaction. Commercially available assay kits expression oxidative biomarkers, superoxide dismutase, malondialdehyde glutathione. In comparison normal cells, model had lower viability, higher apoptosis rates a more severe oxidation status. Melatonin increased viability while lowering stress. It influenced apoptosis-related as well stress indicators. Finally, treatment with able regulate proliferation, cells. The application may be novel strategy protect against age-related changes macular degeneration. has protective effects death. inhibits damage decreases rate.

Extremely low frequency-pulsed electromagnetic fields affect proangiogenic-related gene expression in retinal pigment epithelial cells
Objective(s): It is known that extremely low frequency-pulsed electromagnetic fields (ELF-PEMF) influence multiple cellular and molecular processes. Retinal pigment epithelial (RPE) cells have a significant part in the emergence and pathophysiology of several ocular disorders, such as neovascularization. This study assessed the impact of ELF-PEMF on the proangiogenic features of RPE cells. Mate...
متن کاملEffect of curcumin on aging retinal pigment epithelial cells
Age-related macular degeneration (AMD) is now one of the leading causes of blindness in the elderly population. The antioxidative effects of curcumin on aging retinal pigment epithelial (RPE) cells are still unclear. We conducted an in vitro study to investigate the effects of curcumin on aging RPE cells. A pulsed H2O2 exposure aging model was adopted. Aging RPE cells were treated with curcumin...
متن کاملSimulated microgravity induced damage in human retinal pigment epithelial cells.
PURPOSE The goal of this study was to determine the potential damage to the human retina that may occur from weightlessness during space flight using simulated microgravity. METHODS Human retinal pigment epithelial (hRPE) cells were cultured for 24 h in a National Aeronautics and Space Administration-designed rotating wall bioreactor vessel to mimic the microgravity environment of space. Sing...
متن کاملOxidant-induced apoptosis in cultured human retinal pigment epithelial cells.
PURPOSE To determine the mechanism of oxidant-induced cell death in cultured human retinal pigment epithelium (hRPE). METHODS Cultured hRPE cells were treated with different concentrations of a chemical oxidant, t-butylhydroperoxide (tBH), for different periods of time. Apoptosis was determined with terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick-end labeling (TUNEL) and flow c...
متن کاملCytoskeleton in normal and reactive human retinal pigment epithelial cells.
The cytoskeleton of normal and reactive retinal pigment epithelium (RPE) was analyzed immunohistochemically in five light microscopically normal formalin-fixed, paraffin-embedded human eyes enucleated because of orbital tumor and in 44 eyes with a uveal melanoma. In 26 eyes, the RPE overlying the tumor was morphologically normal or atrophic; in 18 eyes, hyperplastic changes were present. Normal...
